The spleen, as a pivotal immune organ, is responsible for blood filtration, initiation of adaptive immune responses, and maintenance of systemic immune functions. Splenic lymphocytes, comprising B cells, T cells, and NK cells, serve as the core functional units orchestrating antigen-specific immune responses. These cells are isolated through gentle mechanical dissociation of spleen tissue, enzymatic digestion, filtration, and erythrocyte lysis techniques to ensure high viability and purity.
